The SARS‐CoV‐2 Spike Glycoprotein Directly Binds Exogeneous Sialic Acids: A NMR View

چکیده
The interaction of the SARS CoV2 spike glycoprotein with two sialic acid-containing trisaccharides (α2,3 and α2,6 sialyl N-acetyllactosamine) has been demonstrated by NMR. NMR-based distinction between signals those acids in glycans covalently attached to protein belonging exogenous α2,3 N-acetyllactosamine ligands achieved synthesizing uniformly 13C-labelled at acid galactose moieties. STD-1H,13C-HSQC NMR experiments elegantly demonstrate direct residues both additional participation moieties, especially for α2,3-linked analogue. Additional presence a specific antibody N-terminal domain isolated receptor binding domains unambiguously show that site is located domain.

Identification of sialic acid-binding function for the Middle East respiratory syndrome coronavirus spike glycoprotein.
Middle East respiratory syndrome coronavirus (MERS-CoV) targets the epithelial cells of the respiratory tract both in humans and in its natural host, the dromedary camel. Virion attachment to host cells is mediated by 20-nm-long homotrimers of spike envelope protein S. The N-terminal subunit of each S protomer, called S1, folds into four distinct domains designated S1A through S1D Binding of ME...
متن کاملThe biosynthesis of sialic acids.
The sialic acids (1) have in common a 9 carbon carboxylic acid, neuraminic acid (Fig. l), which may be regarded as a condensation product of a 3 carbon and a 6 carbon unit. Neuraminic acid itself is not found naturally, but several Nand O-substituted derivatives are widely distributed in nature (1). Chemical (2-4) and enzymic (5-7) degradations of N-acetylneuraminic acid have shown that this su...
متن کاملDiversity in the sialic acids.
It is now more than 50 years since N-acetyl-neuraminic acid was first discovered and subsequently characterized by several groups (reviewed in Roseman, 1970; Gottschalk, 1972; Rosenberg and Schengrund, 1976; Schauer, 1982; Faillard, 1989). Relatively soon after its discovery, it became apparent that this molecule was actually the major member of a family of compounds related to neuraminic acid ...
متن کاملThe myelin oligodendrocyte glycoprotein directly binds nerve growth factor to modulate central axon circuitry
Myelin oligodendrocyte glycoprotein (MOG) is a central nervous system myelin-specific molecule expressed on the outer lamellae of myelin. To date, the exact function of MOG has remained unknown, with MOG knockout mice displaying normal myelin ultrastructure and no apparent specific phenotype. In this paper, we identify nerve growth factor (NGF) as a binding partner for MOG and demonstrate that ...
متن کاملTerminology for sialic acids.
We write on behalf of the Nomenclature Committee of the International Union of Biochemistry to comment on the proposal by Scott et al. (1982) to change the name of neuraminic acid to amino-sialosonic acid as a consequence of allotting the name sialose to a hypothetical 3-deoxynonulose. They further propose symbolizing sialose as Sia, sialosonic acid as SiaA, and aminosialosonic acid as SiaNA. W...
